~INTENDED USE
Chloramphenicol Rapid Test is a competitive immunoassay for the semi-quantitative detection of the presence of Chloramphenicol in milk.
Cut-off:0.3ppb
Assay Time: 5-10 min
Sample: Defatted milk
KIT COMPONENT
1. Each containing one cassette, one pipette, and a desiccant
2. Diluent
3. Centrifugal tubes and Macrowells
4. Product Manual
TEST PROCEDURE
1. Bring some under tested milk to room temperature (20-30°C).
2. Collect 4mL of milk and 8 mL of ethyl acetate into a centrifugal tube. Shake for 10 min. Do centrifugation at 7000 rpm for 2-3 min.
3. Collect accurately 4 mL of supernatant into a small container. Dry the liquid by blowing wind.
4. Dissolve the residues in the small beaker with 0.3 mL of the provided assay diluent. Stand to make two layers separated.
5. Take out the microwells strip from the plastic canister. Take one well and tear off the film.
6. Use a pipette to collect the lower layer towards the reticle in the pipette (200 μL) and transfer it into the well.
7. Use another pipette to repeatedly suck and extrude the sample until all red reagents are completely dissolved. Wait for about 1 min. NOTE: The sensitivity of the strip is positive correlated to the reaction time. It is s ested to wait for 1-2 min before running the assay.
8. Take out the cassette from the foil pouch and place it horizontally.
9. Suck the mixture in the well and gradually drip 3 drops into the assay sample hole “S”.
10. Interpret the result in 5-10 minutes. Result after 10 minutes is considered as invalid.
NTERPRETATION OF RESULTS
Positive: Only one clear band in C zone indicates a positive result. If a vague T band can be seen but apparently weaker than C band, we also consider it as a positive result. Positive shows that the concentration of Chloramphenicol is above 0.3 ppb in the sample.
Negative: The presence of both clear bands in C zone and T zone. (T band is close to or stronger than C band.)
Invalid: No colored band appears in C zone.
SPECIFICITY
The results are negative when the test card is applied to detect 100 ppm of Aminoglycosides, Tetracyclines, Beta-lactam, Macrolides, Sulfonamides.
STORAGE
All materials must be stored at 2-8°C. The test kit is stable through the expiration date (12 months) marked on the foil pouch. Do not store the test kit in direct sunlight.
PRECAUTION
1. For best results, please strictly adhere to these instructions.
2. All reagents must be at room temperature before running the assay.
3. Do not remove test cassette from its pouch until immediately use.
4. Do not reuse the test kit. Do not use the test beyond its expiration date marked on the foil pouch.
5. The components in this kit have been quality control tested as standard batch unit. Do not mix components from different lots.
LIMITATION
Chloramphenicol Test is a useful tool offering a rapid and accurate testing in field screening, exceeding with its convenience. It provides a semi-quantitative method to detect the Chloramphenicol at or above 0.3 ppb in milk. If you want a quantitative result, it is s ested to apply other methods such as ELISA /HPLC in practice.
